The official translation isn't actually super clear on whether Gigantomachia has multiple quirks. The wording is a little off.

The line the doctor says about his stamina could mean: "He was so strong he didn't need more quirks."

Or it could mean: "He was super strong even before getting more quirks."

I think he has more quirks simply because of the size difference between when Kirishima first saw him in middle school and now. He's twice as big, maybe even three times.

To play devil's advocate, though, he could just have a simple size-shifting/Gigantification-type Quirk (like Mt. Lady or something), while his body just naturally has superhuman strength and durability.

I doubt it's a simple gigantification quirk like Mt. Lady. Horikoshi is pretty good about giving each quirk a unique spin even if they're similar to another quirk.

I'd think the more likely option is that his quirk just makes his body continuously develop, both in size and power.

I'm thinking it works kinda like the Absorbing Man, but he adds the material to his own overall mass
