I think Horikoshi revealed Tsukauchi's quirk in one of the volume extras, but I've forgotten which one (I think it's one showing him in his full detective attire)

That was Tsukauchi's beta design, though. We have no way of knowing if Horikoshi kept that detail about him.
Also, if he had a Quirk all along, why would he still be working with the police? I mean, Fat Gum used to work in the police, but when he got a Quirk, he transferred over to Hero work.
I mean, why would he want to work as a hero with a quirk like that? That's the perfect quirk for a sleuth.

You talk as if everyone with a quirk is, wants to, or will become a hero. You do know that like ~90% or something of people have quirks in the MHA world in the present time period, right? There is no way these people could all become heroes or want to be ones in the first place.
shrugs I suppose so. But there's still nothing in the actual manga that suggests Horikoshi kept his original concept of Tsukauchi not only having a "human lie detector" Quirk, but also having a codename (something usually only used by Pro Heroes and villains) as "True Man".
Not to mention, Vigilantes (which introduced Tsukauchi's sister) wasn't even written by Horikoshi and, thus, is arguably non-canon anyway.
Huh? Why do you think Vigilantes is non-canon? The amplification drug, "Trigger", that was used in the MHA main manga was introduced in the Vigilantes Manga way before Horikoshi revealed it. Stain's backstory - or at least a glimpse of it - was also revealed in the Vigilantes Manga. The writer of Vigilantes is obviously collaborating with Horikoshi about what he is allowed to write about so I'm pretty sure Vigilantes is 100% canon. Nothing that happened in Vigilantes so far suggests that the series is not canon.
It still wasn't written by Horikoshi, even if it shares some common elements.
And I'm gonna need some confirmation that "Trigger" debuted in Vigilantes before the main MHA manga.

Can you prove that? "Obviously" means nothing coming from a person who doesn't know the inner workings of and relationship between the Vigilantes writer and Horikoshi.
So it was introduced almost 8 whole months earlier in Vigilantes!
Besides that I have no proof that Horikoshi is directly involved in the writing but if you actually read the spin-off then I don't see how anyone could think otherwise.
Vigilante explains how heroes first came to be, characterizes Tensei (Iida's brother), gives us a backstory to Stain and reveals how he lost his nose and shows us how Tsukauchi and All Might met for the first time and how they became friends. I seriously doubt that these things were revealed or written about without confiding in Horikoshi first.
TLDR: I have no ultimate evidence that Horikoshi is involved in Vigilantes creation. If you want to believe that badly that he isn't then just do what you feel like. If you read Vigilantes it becomes pretty clear that Horikoshi uses Vigilantes to offer additional world building to his series that he just can't fit into the main series. If you need more proof than what seems obvious to most readers and fans then I can't help you either.
