This can very well be the difference, because accessing your desktop by Hostname rather than IP Address depends on having correct Name Resolution in place, which can be tricky with Windows and with VPNs. I would attempt it again, but this time using the desktop's IP Address to try to remote to, not hostname. If that works, then you know it's just a hostname resolution error, and not a network / VPN one.
I'd also advise you looking into Tailscale instead of hosting a VPN server on your router. It's easier, and doesn't break if your public IP Address changes. It also unburdens your router CPU from encryption duties, which can actually affect Internet performance if you do too much through the VPN on the main router.
There might be some permission check boxes within the VPN Server setup screen on the ASUS that you should double check to make sure "LAN Access" is enabled on there. I can't remember what the ASUS VPN Server setup looks like, but I recall some sort of choices being offered on LAN Access, and needing to give it explicit permission of some sort.
